Human milk oligosaccharides (HMOs) have pronounced antibacterial and antibiofilm properties against Group B <i>Streptococcus </i>(GBS). We have demonstrated that antibiotic combination therapies with HMOs can be effective at increasing antibiotic potency, as well as reversing antibiotic resistance. Untargeted metabolomic analysis has revealed key cell membrane associated metabolites are significantly accumulated upon dosing with HMOs, suggesting a cell membrane dependent mode of action. To better understand the direct bacterial targets of HMOs within GBS, we have synthesized single-entity HMO probes. These compounds feature a variety of core oligosaccharides appended with diazirine and alkyne moieties for use in target identification assays. These probes have retained biological properties and future work will help to identify specific cellular interactions critical in the antibacterial nature of HMOs.
