A new control release system (CRS) of drugs based on pectin and protein in the form of hydrogels and microcapsules in the emulsion is developed. The hydrogel drug delivery form is based on complex coacervation of pectin and corn zein. The microcapsules were tailored from different low methyl-esterified (LM)- and high methyl-esterified (HM)-pectin and B-Lgs concentrate. Both therapeutic systems may withstand pH to protect the active drug through the upper GI system. During 4 hours of CRS administration, the encapsulated drug (piroxicam) was not found in the stomach. The drug concentration was higher in the intestinal and the blood. Drug release kinetics was similar for the Px in tablet and hydrogel (CRS) forms, but with rate constants distinguished in order of magnitude, described by low power Higuchi equation in vitro; at conditions of ex vivo and in vivo, the transport mechanism favors a zero-order drug release rate. From pharmacokinetics data, we found that regardless of the dose administrated, all parameter findings have higher values for the CRS form in comparison with tablet form. Drug release in in vitro and animal experiments demonstrated that the P/Z hydrogel fabricated from LM- and HM- apple pectin are both very suitable as controlled release systems for pharmaceutically active drugs, particularly peptide and proteins, to the colon site. In comparison with the traditional tablet form of Px (model drug), the Px in P/Z hydrogel formulations maintained the drug dose at the colon site for one additional day.
