Glutathione Disulfide (GSSG) plays an important role as an anti-oxidant, including metal-mediated oxidative DNA damage. Iron can generate harmful reactive oxygen species (ROS), such as hydroxyl radical (OH) by the Fenton reaction, and the presence of GSSG may mitigate OH radical production by coordinating iron ions. Oxidative damage by ROS can modify DNA strands, which can lead to numerous diseases and conditions such as diabetes and aging. On the other hand, selective and site-specific ROS generation can be used in chemotherapeutics and are essential for metabolic processes such as homeostasis. Thus, understanding ROS production and monitoring the amount of ROS in the human body is of great importance, which makes understanding the interaction between iron ions and GSSG beneficial. The present study attempts to understand the ability of GSSG to coordinate Fe (III) using Isothermal Titration Calorimetry (ITC). The titration curve shows the stoichiometry of the reaction and can provide thermodynamic quantities, such as enthalpy and entropy for this reaction. These quantities can be ultimately used to find the equilibrium constant for the coordination reaction between iron ions and GSSG.
