Alzheimer’s Disease is a leading form of dementia and affects 10% of adults 65 and older. Barnham et al. (May 13, 2008. PNAS, 105(19): 6813-6818) showed that phenanthroline compounds can bind to β-Amyloid and prevent the formation of plaques, thereby inhibiting the neurotoxicity of Aβ. While this could have major implications for the future of Alzheimer’s treatment and research, a major hurdle still exists since no Platinum (II) 1,10-Phenanthroline products have been documented as water-soluble. This also leads to a gap of information as to the biological reactivity of these water-soluble phenanthroline ligands. The Williams Biochemistry Laboratory is exploring syntheses of water-soluble using Pt(phen)Cl2 and Dichloro-Platinum (II) Neocuproine reagents. The predominant form being explored is the displacement of the halogen by oxalate. Products are being examined by Nuclear Magnetic Resonance. Additionally, reactions with imidazole, histidine, and methionine are being conducted to determine biological reactivity of reagents and products. Future work includes reaction of the water-soluble product with Aβ16,42 to determine whether the adduct inhibits plaque formation, followed in the far future work by in vivo demonstration of the biological activity.
