The exploration of fungal secondary metabolites as a source of, or lead to, new drug discovery is pertinent in the fight against antimicrobial resistance. The wide range of biological activities that these metabolites have been shown to exhibit and the vast potential for isolating new and more potent versions of natural products makes such investigations worthwhile. Secondary metabolites are the end results of activation of the gene clusters that code for their biosynthesis. These genes can be triggered by the manipulation of the environment in which the fungus grows. Paraconiothyrium cyclothyrioides, isolated from the alga Diginea simplex, was grown on solid substrates, such as rice and breakfast cereals. The fermentations led to the isolation of a chromanone, 2-methyl-5-hydroxychroman-4-one, and three 10-membered macrolides – Modiolide A and Stagonolide E, along with a novel polyketide, provisionally named Modiolide C. Modiolide A and Stagonolide E exhibit antimicrobial and phytotoxic activities respectively. The effects of medium manipulation on the production of secondary metabolites is reported.
