Developments in the science of nanotechnology and organic chemistry opened up new avenues in the field of pharmaceutical biology for the innovation of therapeutics ranging from simple nanoparticles to advanced dendrimers that self-arrange themselves into complex organic supramolecular assemblies and called dendrimersomes. Dendrimersomes are complex cell-like nanocarriers that involve different organic reactions to architect amphiphilic Janus dendrimers (JDs) and show attributes to self-assemble in the presence of the aqueous environment into various morphologies usually in spherical shapes and other complex structures. These monodisperse dendrimersomes represent themselves as the mimics of biological cell membranes and can be designated to multifunctional tasks. A library of the 12 amphiphilic Janus dendrimers were synthesized having phenol derivatives (hydrophobic dendron) inspired by plant phenols, an <i>ortho</i>-nitrobenzyl group sensitive to light in the middle of two dendrons and a hydrophilic triethylene glycol chains representing hydrophilic dendron. Each dendrimer in dendrimersomes has hydrophilic and hydrophobic dendrons that align themselves according to their surrounding solvent, molecular structures and the type of hydrophobic side chains into different morphologies were studied by Cryo-TEM. The illumination of the giant dendrimersomes self-assembled by the film hydration method revealed that the single shine of 405 nm wavelength of light under confocal microscopy for 1 s resulted in the cleavage, disassembly, and reassembly into very small vesicles or tubes followed by the incomplete release of the hydrophobic drugs doxorubicin. The cleavage of dendrimersomes into constituent parts may not cause any toxicity to organisms because the design of the dendrimers was inspired by the components that are biocompatible to humans. This study provides a mechanism for the use of photo-responsive dendrimersomes for the incomplete release of cargo and may explain the partial release of drugs as reported in previous studies. Future prospects of this work involve the encapsulation of plant extract synthesized Ag nanoparticles into Janus dendrimersomes and their effects on the release of doxorubicin.
