Guanine quadruplexes (GQs) are structures of DNA or RNA with linear repeats of guanine nucleotides that have the ability to form stacked square planar G-quartets through intra or intermolecular folding. These structures are proposed to have a role in telomere biology, aging, and in the control of gene expression. Various factors influence the folding patterns and topology of G-quadruplexes, including DNA sequence, salt type and concentration, and molecular crowding. Through the use of UV-Visible absorption spectroscopy, CD spectroscopy, and Fluorescence Emission spectroscopy, the conformations of d(TxG4Ty) sequences in the presence of sodium and potassium buffers, and PEG is explored. It is known that dT4G4 and dT4G4T form tetraplex parallel G-quadruplexes. Here we systematically explore the effect of flanking thymine length on either 5’ or 3’ end on the folding patterns and possible interconversions of these quadruplexes.
