With the recent legalization of Hemp Cannabis Sativa nationwide, there has been an increasing demand for phytochemical and cannabinoid analysis. In recent years, the gold and accepted standard for cannabinoid detection and quantification has been performed with High Performance Liquid Chromatography. Although this method has been successful, HPLC requires a large volume of solvents; In addition, standards of the compounds of interest are required along with regular calibration. A quantitative Nuclear Magnetic Resonance (qNMR) spectroscopy approach has been developed for the detection and quantification of cannabinoids such as THC, THCA, CBD, CBDA and CBN. Extraction procedures utilizing deuterated chloroform for immediate qNMR hemp analysis were developed and standardized. The height placement of flower and leaf location on the plant was found to significantly impact cannabinoid concentration. On average, hemp flower located near the top of the plant tested for a 15-20% increase by weight of potential CBD in respect to hemp flower located near the bottom. Monitoring of hemp in the field was undertaken and a substantial increase in cannabinoid concentration during the final two weeks of growth was observed. Cannabinoid concentration in hemp flower and closely associated leaves underwent a 50-100% increase by weight, varying from plant to plant. Decarboxylation of CBDA to CBD was conducted at 105°C, 125°C, and 148°C over the course of an hour. The rate and decarboxylation profile of CBDA to CBD was roughly logarithmic at 105°C, sigmoidal at 125°C, and hyperbolic at 148°C. Finally, a number of topical and oral oil products, vape products, beverages, and edible gummy products were analyzed by 1H qNMR for CBD potency and quality assurance.
