While an arsenal of anticancer and other therapeutic agents has been constructed and employed in treatments, the targeted delivery of these agents as yet to been realized. Nanoparticles (NPs) have provided new avenues in the encapsulation of therapeutic agents, and in some cases do provide targeting arms upon their periphery that aids in their delivery to specific disease states, such as cancer. However, while targeting NPs have allow partial selectivity, they still fail to achieve significant targeted delivery relative to off-target actions. To help correct this, we propose to use an external activating stimulus to allow for a more confined and location specific targeting to be used in combination with NPs and free therapeutic agents. To be presented is our method that utilizes the targeting capabilities of dextrin-coated NPs towards cancerous cells for their enhanced uptake (Stage A), followed by the application of inhomogeneous magnetic pulses (MP, Stage B), which induces microporation (Stage C) that allows for rapid drug uptake by the cell and upon termination of the MP the micropores close trapping the drug within the cell. Doxorubicin (potent anticancer agent) and the lymphoma cancer cell line U937 were employed within this strategy. The transport of doxorubicin into U-937 was increased by 75% when the cells are exposed to few hundred Tesla/m inhomogeneous magnetic pulses. Furthermore, a positive synergistic response was observed within this strategy that could allow for a near 32% decrease in doxorubicin concentration while retaining the same therapeutic efficacy. Validation of the use and targeting potential of NPs and the MP’s will be presented, their lack of individual toxicity towards cells, and the transport of other therapeutic agents into U937, other cancerous and non-cancerous cell lines will also be presented.
