Betulinic acid is a bioactive pentacyclic triterpenoid isolated from the bark of birch trees. Betulinic acid and its derivatives show diverse range of medicinal activity such as their potential applications as anti-tumor, anti-inflammatory, and anti-HIV agents. For the past several years, we have been working on the development of betulin based small molecules as therapeutic agents owing to their importance. Starting from the natural product, we were able to prepare chalcones via aldol condensation, α-acetoxyamides via Passerini reaction and amines via reductive amination reactions. In addition, we have also prepared novel betulinic acid-cinnamamide conjugates using click reaction to couple the two motifs. Some of these conjugates showed significant biological activity as anti-cancer agents. Polyphenols are a class of natural and synthetic organic compounds that show diverse range of antiproliferative, anti-oxidant, and chemopreventive activities. Urolithin A is a secondary metabolite obtained by the conversion of ellagitannins (a type of polyphenol) by gut bacteria. In the present study, we report the second generation synthesis of betulinic acid - urolithin - triazole conjugates in an effort toward understanding the structure activity profiile of our template. The presentation will include our synthetic efforts and biolgical investigation data.
