Prostate cancer is predicted to be the most common, non-skin cancer diagnosed in men in the US during 2019, accounting for nearly 10% of all new cancer cases. However, the false-positive and false negative rates associated with the common PSA test, as well as health risks accompanying follow-up procedures, lead to a need for a novel diagnostic tool. The research presented here intends to develop a screening/early diagnostic tool that utilizes boronic acid functionalized synthetic lectins (SLs) as an approach for the serum-based detection and prognosis of prostate cancer. Our approach is based on the knowledge that during the onset and throughout the progression of numerous diseases, including cancer, aberrant glycosylation occurs which involves the over-, under-, or neo-expression of specific glycans on membrane-bound and secreted glycoproteins. This design presents a unique opportunity for the detection of cancer at the earlier stages of the disease when it is the most treatable as well as providing an opportunity to identify the virulence of the disease. <br/>Initial studies using an array of SLs resulted in a 92% classification accuracy when discriminating human prostate derived cell lines based on metastatic potential. To increase the accuracy of the SL array, new SLs that are selective for prostate cancer are needed.  Using a dual dye, rhodamine and fluorescein, screening method 5 new prostate and prostate cancer selective SLs were identified. These new SLs were able to increase the accuracy of the array when discriminating prostate cells lines based on their metastatic potential to 97%. Specifically, the newly identified SL 11 on average was able to classify the different cell lines nearly 90% of the time. Future work will focus on using serum isolated from pooled blood samples to isolated new SLs that are selective to prostate cancer. <br/>
