Leishmaniasis is a parasitic disease endemic to tropical and subtropical countries. It is caused by the trypanosome Leishmania sp. and transmitted to humans by the insect vector phlebotomine sand fly. Current estimations point to more than 2.1 million cases per year of leishmaniasis infections distributed among the three disease forms: cutaneous, mucous and visceral; being the latter one a life-threatening condition caused by the variant leishmania donovani. Lipophosphoglycan (LPG) is a polysaccharide largely found at the cell surface of Leishmania species. Its intrinsic antigenicity makes this glycan an attractive target for developing carbohydrate-based vaccine candidates and serological diagnostic tools. The structure of LPG consists of a terminal cap saccharide, a repeating unit that builds a phosphopolysaccharide chain and a core glycan that connects the glycan to phosphatidylinositol membrane anchors. In this communication we present our progress towards the synthesis of fragments of the phosphosaccharide chain (repeating unit) for its further development into Virus like particle -conjugated vaccine candidates for visceral leishmaniasis.
