Selenosulfides, when tethered to gem-dimethyl esters, provide an exceptional framework for the development of reaction-based fluorescent probes for H2S detection. In addition to displaying high reactivity and selectivity towards H2S, the distinct chemical differences of selenium and sulfur provides a unique opportunity to enhance the physicochemical properties of these probes without compromising their reactivity towards H2S. In our initial studies, we successfully highlighted this unique flexibility by designing H2S reaction-based fluorescent probes that displayed remarkable biocompatibility and tissue specificity through structural variations of their sulfur linker. With continued modifications that further improve cellular delivery and/or specificity, we predict that this framework will serve as the foundation for a valuable set of chemical probes that may be used to further investigate the biological significance of H2S.
