Nonstructural carbohydrates (NSC) are important for plant growth, development and survival. NSC such as glucose, fructose, sucrose and sugar alcohols are polar molecules that pose unique challenges during extraction and chromatographic separation. Existing methods such as anion-exchange chromatography integrated with pulse amperometric or evaporative light scattering showed low to moderate detection issues. To date, low level detection remains challenging unless a mass spectrometer is used which considerably increases cost of analysis and introduces other issues such as matrix effect. In past few years, hydrophilic interaction liquid chromatography (HILIC) has been evolved and utilized to separate a wide array of polar compounds from complex mixtures. Corona charge aerosol detection (CAD) has become one of the mainstream universal detector that shows a wide linear dynamic range with a significant cost savings for routine characterization. In this study, we developed a HILIC-CAD based methodology using a HILIC-Z (a zwitterionic phase) column to estimate sugars from complex plant matrices such as nectar. Our analysis was carried out in a ThermoFisher 3000 RSLC (rapid separation liquid chromatography) using acetonitrile and aqueous ammonium hydroxide as mobile phase eluents. In our twelve minute analysis, the analytes were retained in the aqueous phase consistently and at the end showed reprducible peak shapes.
