Mucin glycoproteins can regulate cross-kingdom virulence by influencing how microorganisms swim, settle, and communicate. They disarm pathogenic bacteria without inducing resistance, solely disabling virulence factors responsible for disease. The template-independent mechanism of mucin glycosylation and the high-density of complex oligosaccharides preclude a direct investigation of mucin structure-function relationships. Given mucins critical role in shaping host-microbe interactions, understanding the molecular basis of their function is critical and could transform the way we approach infectious diseases. In this project, synthetic analogues of these fascinating molecules have been established and mucin-inspired materials for the domestication of virulent microbial populations have been employed. Two simultaneous approaches for the design of mucin-mimetic polymers will be presented along with proposals for how these materials can be used in microbial toxin inhibition and anti-biofilm assays.
