Understanding the factors that affect the stability ofsupported lipid bilayers (SLBs) on nanoparticles is important for their use inapplications such as drug/DNA delivery. Although both multi (MLV)- and smallunilamellar (SUV) vesicles are stable both in aqueous and buffered media, thesesame lipid bilayers may not be stable when on nanoparticle supports. The fusionof the vesicles onto solid supports is dependent on factors such as the ionicstrength of the media, and the relative size of the lipid to nanoparticle. Inaddition, the amount of lipid needed to form a supported bilayer may depend onthe nanoparticle size. In order to understand these effects, the fusion andstability of dimyristoyl-, dipalmitoyl-, and distearoyl- phosphatidyl choline(DMPC, DPPC and DSPC) SUVs onto silica nanobeads with nominal diameters ofbetween 5 and 100nm has been investigated using dynamic light scattering, zetapotential measurements and nano-differential scanning calorimetry.
