Competitive 1,1'-oxalyldiimidazole (ODI) chemiluminescent enzyme immunoassay (CLEIA) for the rapid screening of melamine in dairy foods was developed. Melamine (150 �l) in 20 % (milk: PBS at pH 7.4 = 1:4) and 100 % milk was incubated withmelamine-conjugated horseradish peroxidase (50 �l) in a strip-well coated withrabbit anti-melamine for 20 minutes at 37 �C. After the washing thestrip-plate using PBS containing 0.05 % tween-20, 100 �l of Amplex Red (25 �M)containing 0.4 mM H2O2 was inserted into the strip-wells.Resorufin formed in each strip-well during 5 minutes of incubation at roomtemperature was quantified with ODI-CL detection. ODI-CLEIA (dynamic range:3.8-250 ppb) using 20 % milk was at least 50 times more sensitive thancommercially available EIAs. ODI-CLEIA (dynamic range: 125-2000 ppb) capable ofdirectly quantifying 100 % milk without any pretreatment was two times fasterthan ODI-CLEIA using 20 % milk.
