Electrospray Ionization (ESI) has enjoyed a rich history in the investigation of noncovalent interactions by mass spectrometry (MS). It offers exceptional speed, sensitivity, and direct access to interaction stoichiometry without the need for labeling or immobilization strategies. However, MS-determined binding constants still require validation by established solution phase binding techniques due to potential disparities in the ionization responses of monitored species. We have been focused on developing novel, rapid, and reliable flow injection analysis methods for MS-based binding determinations. With a significant focus on small-molecule interaction systems, we must constantly consider the effect of ionization response on our results. This talk will present the methods our group has developed, especially highlighting a new method based on a continuous flow stirred vessel (CFSV), which we believe can remove the response dependence of results and ideally, promote ESI-MS as a more reliable stand-alone routine binding determination technique.
