Alzheimer's disease (AD) is the most common neurodegenerative disease. The brains of AD patients are characterized by the deposition of amyloid plaques that contain the amyloid b (Ab) peptide. Abnormal interactions of Cu and Zn ions with the A� peptide are proposed to play an important role in the pathogenesis of AD, and disruption of these metal�peptide interactions using chemical agents holds considerable promise as a therapeutic strategy. Herein we will present the synthesis and characterization of bifunctional compounds that have high affinity for both Ab aggregates and metal ions, as well as their effect on the neurotoxicity of the various Ab species. In addition, we will present current studies focused on the design of related bifunctional chelators as imaging agents for the detection of Ab aggregates in vivo.
