Complex macromolecular ligands are often needed to test hypotheses related to lanthanide chemistry in magnetic resonance imaging and luminescence imaging. However, these ligands can be difficult to synthesize because of unwanted byproducts, long reaction times, and tedious purifications. To aid in the synthesis and purification of macromolecular ligands, we have used solid-phase and microwave-assisted synthetic techniques. Here, we present the use of these techniques towards the synthesis of cryptands and large polyaminopolycarboyxlate ligands. Our solid-phase and microwave-assisted protocols lead to shortened reaction times and minimization of the formation of unwanted byproducts. We expect that the ability to synthesize macromolecular ligands on solid-phase will enable exploration of exciting frontiers in lanthanide chemistry.
