Fluorescein is a common fluorescent probe used in the biosciences. The high quantum yield and overlap of the excitation wavelength with the Ar ion laser makes it an excellent probe for the biosciences.  However, fluorescein undergoes several solvent dependent transitions that make it problematic for use in nonpolar solvents.  Moreover, fluorescein is especially problematic to use in the lipid bilayer.     In this poster we will present fluorescent and UV-VIS characterization of fluorescein in mixed solvent system of varying pH values.  A comparison will be made between commercially available hydrophobic fluorescein derivatives and fluorescein.  Finally, the Raman spectra of the hydrophobic fluorescein derivative will be compared with that of the fluorescein molecule on hydrophobic glass surfaces.
