We describe two novel assays (GIGI and GIGT) that facilitate the quantitative analysis of the trafficking of glucocorticoid-tagged proteins and peptides from endosomes into the cytoplasm of live cells. By utilizing high-affinity �super� glucocorticoid receptor (GR*) fusions in concert with eGFP expression or translocation, these techniques offer dramatic improvements in speed, cost, and versatility and can be executed in high-throughput. Together, GIGI and GIGT provide means to delineate structural determinants that guide intracellular trafficking of proteins, peptides and mimetics thereof.
