To circumvent the often tedious modification of novel bi- or polydentate ligands, a modular and supramolecular approach might be a potential alternative: Two monodentate ligands interact with each other in a well-defined manner, e.g. one being a hydrogen bond acceptor (A-Type), the other a hydrogen bond donor (D-Type), resulting in a quasi bidentate ligand. In this manner, a large number of different ligands are accessible by simply combining different monodentate ligands with each other.Some 500 catalytic asymmetric hydrogenation reactions were performed to screen the different ligand-ligand combinations. With the right combination of donor-acceptor ligand units, an amplification of enantioselectivity of such hetero-bidentate ligands relative to the homo-bidentate ligands was observed. Use of Chemspeed's CATSCREEN 96 allows for the parallel, automated synthesis of such ligand building blocks and for the parallel, automated screening of the resulting catalyst library.
