Recently, nitric oxide (NO) has been shown to be a potent anti-platelet and antimicrobial agent that can prevent thrombus formation and reduce bacterial adhesion. In this study, poly(lactic-co-glycolic acid) (PLGA) is used along with diazeniumdiolate in a polymer matrix to sustain the NO release for more than 14 days (by maintaining the pH inside the polymer coating). These coatings are evaluated within short-term (4h) and long-term (>10d) in vivo rabbit models of thrombogenicity and infection. In the short-term model, NO releasing coatings were able to decrease the platelet activation and clot formation as compared to the controls while maintaining the platelet function. In the long-term model, the NO release catheters showed a significant decrease in the thrombus and biofilm formation as compared to the controls.
