Naturally evolved RNA structures have been discovered that bind specific small molecules or proteins and regulate gene expression downstream of that recognition event. We have found that these highly conserved RNA aptamers serve as privileged scaffolds that are readily adapted for �unnatural� functions inside cells. In particular, our lab is focused on applying RNA aptamers toward building complex gene regulatory circuits, developing fluorescent biosensors, and expanding the reaction scope of enzymes. Here we will present the most recent developments in these projects, including the generation of RNA-based fluorescent biosensors for cyclic dinucleotides, an expanding class of signaling molecules with dual roles in bacterial pathogenesis and mammalian innate immunity.
