Cation exchange chromatography (CEX) is widely used in the purification of monoclonal antibodies to remove product aggregates, host cell proteins, and DNA.  Previous studies have shown that CEX chromatography can also provide virus clearance, specifically removal of the model virus xMuLV.  The magnitude of virus removal and the mechanism of separation, more retention to the resin relative to the product during elution, are generally understood in relation to such operational parameters as pH and elution ionic strength.  However, the importance of the CEX resin characteristics on xMuLV binding is unknown.  In order to better understand the interaction of xMuLV with CEX resin, we examined the impact of four resin characteristics on the adsorption and elution of xMuLV.  The results of these experiments will be presented.
