Triazole compounds play a key role as antifungals in agriculture and in human mycoses and as non-steroidal anti-oestrogens in the treatment of oestrogen-responsive breast tumours in postmenopausal women. At high doses, these compounds can affect the liver, the thyroids, reproductive organs, fertility, and development in several species. There is increasing evidence for adverse effects of high doses of triazole fungicides on the mammalian steroid metabolism. This review will cover the molecular initiating events which will involve looking at chemical properties of triazoles, macro-molecular interactions (receptor-ligand interactions) with sterol 14a-demethylase and/or aromatase. Cellular responses such as gene activation/silencing, protein production, protein activation/inhibition, effects on inhibition of certain pathways of steroidogenesis will also be considered.
