We have selectively detected integrin av�3 in the membrane of a cultured cell by binding an RGD-functionalized nanoparticle to the integrin and detecting the interaction via tip enhanced Raman scattering (TERS). Far field imaging experiments show that the Raman spectrum observed from just the nanoparticle contains mostly ill-defined bands. While the Raman spectrum observed from the TERS detection show bands that are directly attributable to the targeted protein. This approach appears to overcome challenges common to TERS imaging, specifically locating the protein on the vast membrane landscape and discriminating the protein of interest from the surrounding biomolecules. Given the role of integrins in cell adhesion and differentiation, our studies may provide new insight into the chemistry of these important receptors. In this presentation we will discuss our methodology and its application to the integrin-RGD system.
