Increasingly, a number of new active pharmaceutical ingredients (API) under development display limited aqueous solubility resulting in slow or incomplete dissolution, low bioavailability and poor pharmacokinetics. Lacking further pharmaceutical development, the properties of these drug candidates would critically limit their absorption and bio-performance when administered orally. As a result, a fundamental knowledge of the physicochemical properties of the API is essential in designing strategies to improve solubility/dissolution rate via formulation development leading to new and effective therapies.The permeability and solubility classification and the factors that affect oral absorption and dissolution will be highlighted. General strategies to enhance solubility and pharmaceutical developability will be presented. These approaches typically include particle size reduction, non-crystalline/amorphous forms, solubilized formulations, and crystal modifications. The strategy discussions will incorporate fundamental understanding of the physicochemical properties of the formulation with case studies.
