The production of commodity chemicals from renewable materials is appealing as these strategies are not directly influenced by the cost or availability of fossil fuel resources, and often also benefit from a reduction in organic, inorganic and greenhouse gas wastes associated with formation of these same compounds from petroleum distillates. �-carboxy-cis,cis-muconic acid, which is a metabolic intermediate in the catabolism of phenolic compounds by some bacteria, has been recognized as a novel polymer precursor. We have constructed a de novo biosynthetic pathway to �-carboxymuconic acid comprised of genes encoding enzymes associated with catabolism of aromatic compounds (dihydroxybenzoate dioxygenase) and siderophore biosynthesis (dehydroshikimate dehydratase). Expression of this pathway from a multi-monocistronic plasmid construct in an E. coli host was employed to form �-carboxymuconic acid from glucose via the shikimate pathway. Further, conversion of �-carboxymuconic acid to 3-carboxymuconolactone, which itself has applications in the industrial synthesis of polyurethanes, was achieved via a facile chemical conversion from the triacid. Product isolation and chemical characterization will be discussed.
