Aptamers are short single-stranded DNA or RNA oligonucleotides, which can be selected from synthetic random oligonucleotides library by SELEX (systematic evolution of ligands by exponential enrichment) in vitro,   and they have high binding affinity and specificity for their targets. Aptasensors are a new kind of biosensors based on aptamers as biorecognition elements, and they have been broadly applied in the detection of all kinds of targets. Selection of aptamers against HBsAg would contribute to the better detection and diagnosis of hepatitis B virus infection. In this study, we first selected DNA aptamers against HBsAg by immobilizing HBsAg on the surface of carboxylated magnetic nanoparticles (MNPs), which is convenient for rapid magnetic separation. After 13 rounds selection, DNA aptamers that specifically bind to HBsAg were successfully obtained, and the secondary structures of aptamers were analyzed by DNAMAN software. Furthermore, we have developed aptasensors based on nanoparticles and chemiluminescence to quickly and efficiently detect HBsAg, which may find wide applications in the field of detection.
