The methods that popularly used to detect pesticide residues are based on High Performance Liquid Chromatography (HPLC) and gas chromatography with electron capture detector (GC-EDC). These methods seem to be inconvenient and unaffordable to the farmer or province laboratory agencies due to high cost and complicated. Recently, immunological technique such as  Enzyme linked immunoassay (ELISA) provide a simple, rapid, reliable and inexpensive mean of detection for semi-quantitative assessment of pesticide residues.A fast and simple test was developed based on ELISA technique to detect endosunfan and other pesticides belong to cyclodien group. Polyclonal antisera were raised in rabbits against immunogen developed by conjugating protein to endonsunfan hapten using an activated ester method through a spacer arm attachement. Endosunfan hapten were synthesis from reaction between 1-hydroxy chlordane and succinic anhydride. A direct competitive ELISA was designed  to detect the sensitivity and selectivity of the antibody. All the factors affecting ELISA kit were optimized: antibody dilution is 1�g/well, enzyme conjugate dilution is 0.02 �g/ml, pH 7.4. This ELISA kit show the high sensitivity with endosunfan (LOD = 0.5 ppb, IC50 = 1.0 ppb). More over, it can detect almost cyclodien pesticide group: Endosulfan sulfate (IC50 = 0.6 ppb), Aldrin (0.6 ppb), Dieldrin (0.6ppb), Chlordan (9.6 ppb), Heptachlor (0.1ppb), Endrin (5.3 ppb) and Lindane (68 ppb).
