Five million Americans suffer from Alzheimer's disease (AD); 1 million from Parkinson's (PD); 400,000 from multiple sclerosis (MS); 30,000 from amyotrophic lateral sclerosis (ALS or Lou Gehrig's disease), and 30,000 from Huntington's disease. Because neurodegenerative diseases strike primarily in mid-to late-life, the incidence is expected to soar as the population ages. These Neurodegenerative diseases are characterized by neuronal loss in specific brain regions and by corresponding clinical symptoms. Finding treatments and cures for neurodegenerative diseases is a goal of increasing urgency. For our ongoing chemical biology projects, to study retinoic acid (RA) signaling pathway, we developed receptors specific agonist and antagonist using our LRD (Limited Rational Design) approach and tested their biological activity as potential therapeutic agents for diseases modulated by RA pathways giving emphasis to neurodegenerative diseases AD.Vitamin A and it's metabolites called all trans-retinoic acid (atRA) is known as Retinoic acid alpha agonist has been shown to have a beneficial effect on mouse models of AD through the up-regulation of the a-secretase enzyme resulting in a reduction of the toxic amyloid-� load in the brains. In this talk, I will discuss detail the synthesis strategy and screening technology we used for our study to develop novel therapeutic agents for the treatment of AD, PD and other NDDs targeting RA signaling pathways.
