Nitrosative stress mediated Parkinson's disease can be linked to blockage of reduced cysteine amino acid of protein disulfide isomerase with NO radical. Different environmental factors can contribute towards this form of Lewy body disease. We followed rotenone mediated mitochondrial dysfunction model to demonstrate protective characteristic of polyphenolic phytochemical, ellagic acid against rotenone insult in PC12 cell line. Our data revealed the anti-apoptotic, antioxidant and anti-S-nitrosylation capabilities of ellagic acid when used against rotenone insult.  In this study, we also have monitored the aggregation of green-fluorescent protein (GFP)-tagged synphilin-1 and a-synuclein (a Parkinsonin biomarker) as a function of rotenone insult. We report that the polyphenolic phytochemical, ellagic acid reduces markedly the incidence of synphilin-1and a-synuclein aggregation if pre-treated for six hours before rotenone exposure. We further reporting that ellagic acid can reduce the Lewy body like aggregation, mimicked by co-expression of synphilin-1 and a-synuclein in PC12 cell.    Our work is a strong indication towards the prophylactic abilities of ellagic acid and can be a scaffold molecule against free radical induced neuronal stress.
