I will dicuss criteria for designing/mutating natural protein sequences to produce extremely stable and fast folding proteins. In the extreme case, these proteins fold without free energy barriers, and have stabilities 10s of kJ/mole greater than typical mesophiles, and even thermophiles in some cases. Protein evolution for function and compatibility with the cellular milieu can frustrate folding speed and stability.
