Source and detector technology has fueled tremendous advances in optical measurement technology over the past 30 years.  I will introduce our current work on neuroscience measurement tools by recalling the technology that allowed the advances in optical measurements that makes single molecule imaging a pipe dream in 1993, and routine practice 2013.  Following this introduction, the daunting measurement challenge of understanding brain function will be reviewed with examples of our work determining anatomy in sectioned brain tissue, anatomy in whole Drosophila fixed brains, and neural activity with new fluorescence sensors, extracellular and patch electrophysiology.
