Quantum dots (QDs) are a relatively new class of fluorophores that are primarily synthesized with a cadmium selenide core surrounded by a capping polymer. In pursuit of a less toxic and more biologically relevant QD formulation, we developed a peptide sequence capable of mineralizing zinc sulfide QDs in a single, aqueous step. This peptide has been conjugated to several proteins and retains its functionality while also allowing for secondary functionality through engineering of the protein. We investigated potential routes of toxicity for the QDs including <i>E. coli </i>cell penetration, DNA damage, protein misfolding, or direct oxidative stress. In order to improve the stability of the QDs without the use of a polymer shell, the fusion protein was derivatized with a silica-binding peptide on the opposite side of the protein allowing for encasement within silica nanoparticles or use within silica gels. Encapsulation of the QDs yields opportunities for sensing or other operations in harsh conditions that would be otherwise inaccessible. These multifunctional proteins provide a toolbox for further development at the organic-inorganic interface.
