With the desire to continuously provide solutions to improve the purification of biomolecules, Pall is about to launch two new generation sorbents: One cation exchange and one cationic mixed-mode sorbent. These sorbents were designed to provide novel selectivities associated with high productivity processes. Characterization of the properties of the CEX sorbent highlights a high capacity maintained over a wide range of pH and conductivities. This feature along with low elution volumes allows for the development of high productivity steps compared to leading currently available sorbents, while maintaining a competitive level of impurity clearance. On the other hand, the associated mixed-mode sorbent was specifically designed with a unique selectivity compared to previously available resins. The sorbent displays improved separation performance compared to commonly used mixed-mode resins as it retains the ability to separate proteins with similar pI and/or hydrophobicity over a wide range of conductivities. Importantly evaluation of regeneration capability of the mixed-mode sorbent demonstrated stable binding and elution performance over repeated cycles using industrial mAb feedstock. The present work shows through process examples how these sorbents can help resolve purification challenges for a variety of biomolecules.
