We have recently developed a novel DNA/drug co-delivery system that requires no carrier materials<b>.</b> In this system, the drug and the nucleic acids themselves <i>are</i> the delivery vehicles. By connecting the drug and the DNA via a “smart” linker to form an amphiphile, micellar structures can be formed. These micelles are capable of cell entry and cellular gene regulation in the absence of a transfection agent. Another unique feature of this system is controlled release of payload. Upon exposure to an appropriate environmental trigger, it is capable of disintegrating into free nucleic acids and releasing payload chemotherapeutics.
