In this study, we designed a conjugation of DNA hairpin structure to the poly(ethylene glycol) molecule to enhance the temporal resolution of low molecular weight poly(ethylene glycol) in nanopore detection. This design makes it possible for detection of an individual PEG with molecular weight as low as 140 Da at single-molecule level in solution. Current fluctuate across nanopore with pico-amp resolution was measured and recorded by home-made multi-channel amplifier system. This novel strategy could also be applied in other characterization of small molecule based on nanopore. Furthermore, we found that the current duration time of poly(ethylene glycol) was scaled with the relative molecular weight, which has a potential application in single- molecule detection.
