In this presentation I examine protein selectivity in a range of chromatographic systems including ion exchange, hydrophobic interaction, hydroxyapatite, multimodal and peptide affinity systems. For each system, a number of biophysics and modeling tools will be employed in concert with high throughput chromatographic data using targeted protein libraries to establish the key determinants of protein selectivity. Important protein surface properties that strongly affect selectivity will be identified for each chromatographic system. In addition, predictive modeling tools such as quantitative structure activity relationship (QSAR) models and coarse-grained approaches will be presented for a number of systems. Finally, the application of these approaches to complex biological products such as Fabs and bispecific antibodies will be presented.
