The non-mevalonate pathway is responsible for the synthesis of isoprenoids in plants, parasites, and in many bacteria. One of the enzymes involved in this pathway is a zinc metalloenzyme called 2-methylerythritol 2,4-cyclodiphosphate synthase (IspF). This metalloenzyme is an attractive target for the development of novel antibacterial agents. In this study, we have used a known selective zinc-binding group to target the active site of IspF in order to develop a new series of IspF inhibitors.
