The colchicine binding site have been a target for tubulin polymerization destabilizers and the development of cancer pharmacophore approaches. Several molecules interacting with the colchicine binding site have been intended and synthesized with significant structural diversity. Combretastatin A-4 and its derivatives have been some of the noted scaffolds for the development of cancer pharmacophore designs. Researchers have been persuasive during decades with the task to establish the moieties with biological potential. When looking forward for choices for therapeutic processes, chalcone-based compounds have been highlighted candidates for the treatment of cancer, malaria, and bacteria. Furthermore, incorporating ferrocene in the design of products, have been covered in the literature with enhancements in the potential of bioactive compounds. Moreover, the combination with heterocycles as an improvement to the compound core, have been guiding researchers in tactics for a plausible heightening in biological potential. Among others, isoxazoles and epoxides have been renowned in the literature as frameworks with analgesic, anti-inflammatory, and anticancer potential. Our synthetic approach foresees these aspects, imposing and emphasizing ferrocenyl chalcones and the incorporation of heterocycles to their structure. To achieve our specific aim, molecular modeling studies have been performed to the ferrocenyl derivatives to inquire properties that would lead us to a pharmacophore design. Moreover, ferrocenyl chalcones have been synthesized via Claisen–Schmidt condensation in moderate to good yields. Additionally, precursors for epoxide products have been accomplished in moderate to good yields aiming for a Darzen-type reaction for the synthesis of epoxide derivatives. Besides, isoxazole compounds have been synthesized in moderate yields from ferrocenyl chalcones. The synthetic products have been characterized by 1H and 13C NMR spectra, cyclic voltammetry, and UV/VIS spectra. Synthetic methodology, characterization, and preliminary bioassays in cancer cell lines, bacteria, and malaria of these compounds will be elaborated.
