Currently, most fluorescent techniques mainly involved the irreversible reaction of a non- fluorescent probe molecule with a free radical of interest to produce a detectable fluorescent product. Such “one-way” irreversible detection methodologies can have limitations, as they are not able to respond to dynamic changes to the cellular redox environment. Because the chemistry of those probes response is irreversible, most techniques therefore cannot be used to identify the potential therapeutic benefit of antioxidant intervention following the induction of a pro-oxidant stress. As “two-way” reversible probes, MitoProbes we developed can be used for real-time monitoring the dynamics of mtROS production continuously in vivo. We have successfully demonstrated their application for the detection of mitochondrial oxidative stress in different oxidative stress-mediated diseases models. <br/>
