Glioblastoma multiforme (GBM) is the most malignant brain tumor due to invasive growth. The migratory tumor cells penetrating normal parenchyma prevents complete surgical removal, while the high resistance to radio- and chemotherapy further result in tumor recurrence with more invasive phenotypes. Although it is well established that the degree of surgical resection directly correlated to patient survival, most surgery is performed without any image guidance. Near-infrared fluorescent probes (NIRF) are being developed for <i>in vivo</i> optical imaging technologies as convenient alternatives to those comprehensive imaging modalities in preclinical studies for tumor detection, and are showing promising results in clinical image-guided oncological surgery. In this study, we evaluated two formulations, liposomes and phospholipid micelles to incorporate IR780 fluorescent dye for <i>in vivo</i> brain tumor imaging using a human GBM orthotopic xenograft model and a spontaneous mouse GBM model. Comparing with IR780-liposome, IR780-micelles showed enhanced intra-tumoral accumulation in both tumor models. Moreover, IR780-micelles also showed preferred intracranial tumor accumulation and potent NIRF signal intensity in GBM spontaneous models at a real-time, non-invasive manner. We conclude that IR780-micelles demonstrated tumor-specific NIRF imaging capacity in glioma preclinical mouse models, providing great potential for clinical imaging and image-guided surgery of brain tumors.
