Indocyanine green (ICG) is the only near infrared (NIR) fluorescent agent for clinical use approved by the European Medicines Agency (EMA) and the U.S. Food and Drug Administration (FDA). However, some of its drawbacks, such as easy aggregation, thermal degradation, photobleaching and rapid blood clearance, have greatly hindered its further application. In this work, we reported that DSPE-PEG could induce the formation of ICG J-aggregates via simply stirring the aqueous mixture of DSPE-PEG and ICG in room temperature, and formed DSPE-PEG@ICG J-aggregates composite nanoparticles (DPIJ NPs), which was dispersed well in aqueous solution with a good monodispersity, and showed excellent photo- and thermal-stability in vitro and in vivo, as well as outstanding photo-thermal conversion capability. The photo-thermal therapy (PTT) and photoacoustic imaging (PAI) ability of the DPIJ NPs were determined in vivo. Compared with free ICG, the obvious enhanced permeability and retention (EPR) effect, the longer absorption maxima and the higher photo-thermal conversion efficiency make the DPIJ NPs much more efficient in tumor ablation and photoacoustic imaging.
