In this talk I discuss the synthesis and application of acyclic cucurbit[n]uril type receptors (e.g. 1 and 2) and clickable monofunctionalized CB[7] (3). Specifically, I will present the results of our structure activity relationships (e.g. influence of glycoluril oligomer, aromatic wall, solubilizing group) on the use of acyclic CB[n] as solubilizing excipients both in vitro and in vivo. Subsequently, I will discuss the use of acyclic CB[n] as in vivo reversal agents for the neuromuscular blocking agents rocuronium, vecuronium and cisatracurium and our recent results on the reversal of hyperlocomotion of rats induced by methamphetamine. Lastly, the strategic merger of the outstanding structural features of metal organic polyhedra with the outstanding recognition properties of cucurbiturils and their use in drug delivery applications will be discussed.
