The glycosaminoglycan (GAG) family of polysaccharides includes the unsulfated hyaluronan and sulfated heparin, heparan sulfate, keratan sulfate, and chondroitin/dermatan sulfate. GAGs play structural roles in connective tissues, tethering cell surfaces to protein and proteoglycan molecules through interactions with lectin domains. Understanding the structure of GAGs can promote further understanding of their biological functions. GAGs possess considerable heterogeneity in average molecular mass, molecular mass range, disaccharide composition and content, and position, of sulfo groups. Capillary isoelectric focusing (CIEF) method is a separation method, which is based on the analyte’s isoelectric point (pI) in the presence of amphlyte. CIEF is widely used in the characterization of proteins and antibodies with high performance. However, this method has never been used to characterize negatively charged GAGs due to their low pI values. In this novel reverse polarity capillary isoelectric focusing method, different types of GAGs can be focused at different PH zones in the capillary. Therefore they are separated and characterized by a negative ion mode mass spectrum.  <br/>
