Terpenes are compounds naturally present in plants and predominantly in essential oils, widely relevant for aroma, biofuels and pharmaceutical industries. Biotechnological production of these terpenes as alternative to extraction from natural sources or chemical synthesis has been encouraged to overcome seasonal supply, low yields and customer rejection to artificial labels. A recombinant <i>Escherichia coli</i> strain maximum concentration (43.1 g biomass/kg broth) produced 0.40 g sesquiterpene/kg broth at age 84.3 h through mevalonate pathway. Viable cells determination stability along time constrasted to optical density profile. Dodecane was used as organic phase for in situ product recovery of sesquiterpenes, compounds often toxic to biocatalyst cells. The growth rate feed phase started at 0.16 h-1 and presented a decreasing profile, typical of constant feed experiments. The substrate used was glycerol, a highly available by-product from the biodiesel industry, in persperctive to encourage economic attractiveness. Higher than 1g/kg terpene has been aimed to allow more feasibility to the process.
