The objective of this research was to synthesize and characterize manganese carbon monoxide-releasing molecules (CORMs) with thiosemicarbazone ligands. In the past 20 years, CO has proven to be an important signaling molecule, with involvement in muscle relaxation, vasodilation and blood pressure regulation. Although large doses of CO can be potentially toxic, it can also exhibit proapoptotic effects in some dysregulated hyperproliferative cells such as cancer cells. CO release has been shown to be useful as an anti-inflammatory, anti-microbial, and cytoprotective agent. CORMs have been investigated as complexes that can release controlled doses of CO to targets in the body, and research shows that metal carbonyls have great potential to be CO-releasing. In this study, manganese CORMs were synthesized with a series of ferrocenyl thiosemicarbazone (TSC) ligands which varied in their functional groups attached to the amine. The synthesized product followed the formula [Mn(CO)3(TSC)Br] where TSC = AcF-RTSC or Fc-RTSC and R = H, Me, or Ph (and AcF being acetylferrocene and Fc being ferrocene carboxaldehyde). Characterization of the synthesized complexes was done using elemental analysis, NMR, infrared, cyclic voltammetry and electronic spectroscopy. The CO-releasing properties of the complexes were determined using the myoglobin assay, and the antibacterial potential of the synthesized complexes were investigated using various types of pathogens. The synthesized complexes inhibited the growth of E. faecalis, P. aeruginosa, K. pneumoniae, and Strep. pyogenes at concentrations ranging from 0.05- 1.00 mM. The complexes are also photo-CORMs that release CO when irradiated with light of wavelengths 365 nm, 405 nm, and 435 nm. The time for CO emission from these complexes ranged from a few minutes to over 5 hours.
